Peter Parker is the alter ego of the web slinging Spiderman. It is through Peter that the spirit of these spiderman comics touches the hearts of so many fans.
How Peter Parker got his powers
Peter Parker is a sensitive guy. He was a Science geek. He was weaker than the other guys, needed glasses to see clearly and was afraid of heights. Often picked on by school bullies. A brilliant, shy, sincere boy who kept to himself.All that changed one day when a spider bit him. In the Spiderman movie, he went on a school trip. A genetically enhanced spider had escaped. When everyone was gone, and Peter was distracted by his feelings for Mary Jane, that spider bit him, making him feel very sick. He goes home, passes out in his room and wakes up all buffed up with the proportionate strength of a spider. He could see clearly without his glasses. When the bullies attack him, moved way to fast for them and hit a bully so hard, he went flying. A delightful, very satisfying action sequence in the Spider-man movie. He finds he can outrun a bus, shoot webbing from his wrists and even climb walls.
In the Amazing Spiderman, he got his powers the same way, except that the spider was a radioactive one, rather than a bio-engineered spider. Right after the spider bite, Peter was unnerve and left the building. He was almost hit by a car but jumped out of the way and found himself stuck high up on a wall. Terrified, he gripped a metal pipe so hard, it crumpled like paper in his hand. He fell and landed on his feed.
In the Ultimate Spiderman, the spider had been injected with a drug called Oz, developed by the Norman Osborne's company. That drugged spider bit him. he fainted momentarily, out of shock. After that, he went back in the school bus, with Mary Jane, uneventfully. Norman Osborne had his people keep an eye on Peter. They paid for Peter's medical bills and gave his a gift basket hoping the Parkers would not sue them. Uncle Ben decided that it was an accident and Harry Osborne is one of Peter's very few pals, so they let it be. The next day Kong, the bully aims a running kick at Peter, who suddenly turned and flipped him. Then Peter passed out again. The Oscorp studied Peter's blood and thought the drug was killing him. To protect himself, Norman sent an assassin after Peter, to kill him a car accident. Peter leaped over the car, increasing Norman's interest in his condition.

Why Peter Parker became Spiderman
As for how Peter became Spiderman. Both the movie and the Spiderman comics tell the same story. He tried to win some money by taking up a challenge in a wrestling ring. He wins, but the organizer refuses to pay up. The organizer gets robbed and the thief runs off, heading in Peter's direction. Peter lets the thief get away, and the thief shoots uncle Ben who tries to stop him later. Out of guilt, in memory of Uncle Ben, Peter uses his powers to fight crime, as Spiderman. Uncle Ben's words 'With great power comes great responsibility' drive him to do what he must as Spiderman, so that no one else will get hurt because of his inaction.

Peter Parker's love life
Before he got his powers, Peter was unlucky with girls. In Ultimate Spiderman, Mary Jane was already his friend before the bite. But in the movie, she barely noticed his existence.After he got his powers, Peter became more confident with girls. In the spiderman comics, he dated Mary Jane, went steady with the beautiful Gwen Stacy, he went out with Betty Bryant. Even the black cat, Felicia Hardy fell for him, or rather, for his alter ego Spiderman. He became quite a ladies men.
Still, nothing went right for him. Peter can hardly keep appointments with his girlfriend. He is constantly fearful of her safety. All the wacko villian s who know his secret identity make a beeline for his girl.
In Spider-man, Green goblin kidnaps Mary Jane, to force Spiderman to choose to save her or the children as he drops them to their death. Spiderman saves them both. In Spider-man 2, Doc Ock kidnaps Mary Jane (again) to force Peter to bring Spiderman to him. All that makes Peter afraid for Mary Jane's safety. So despite his feelings for her, he avoids having a relationship with her. In the Spider-man 2 trailer, when she asks him directly if he loves her, he says: I don't.
In the spiderman comics, one of the most memorable stories was the one where green goblin kidnaps Gwen Stacy to lash out at Spiderman. He drops her to her death. Spiderman catches her with his web, but her back snaps. She dies instantly. So ends to beautiful love story between Peter and Gwen.
In the Ultimate spiderman, The Green Goblin (again.. it always has to be him?) kidnaps Mary Jane. He drops her to her death but Peter catches her just in time. That incident shook Peter up, but Mary survived.
Peter Parker's family
Peter's only family is his Aunt May. She plays a key role in his life. In Spider-man 2, it was her conversation with him, about heros, that got him to want to be Spiderman again. His parents had died when he was a baby. His Uncle Ben and Aunt May raised him as if he were their only child. They were like his parents to him. When Ben died, Aunt May was all the family he had.Peter Parker's problems
Spiderman strikes a chord with us because of his very human problems. Money problems. Finding it hard to get money to pay the rent. Relationship problems. All these are things we can identify with.He uses his powers to help others, and make some money by peddling photos of himself in action. Something very human too.
Sometimes the sacrifices he makes because of his life as spiderman are so great that he has toyed with the idea of giving it up. Something that happened in Spider-man 2 and in the Spiderman comics.
He has long conversations with himself. Pondering over his problems, so much so that sometimes, I think he needs to see a psychiatrist.
All these, make Peter Parker a very endearing character.
